Unfortunately no. as an alternative to being able to change the default palette, you can save a .pyxel file with a different palette setup, and the use that as the base file for your spriting.
My own workaround for not being able to add more presets: make a folder named "palettes" that is easy to access in path, then use Color->Import from image
