It could be that Pyxel Edit checks for the actual key press, and not just if the modifier is held when clicking. I'll make a note to investigate.
Wacam Intuos Pro on OSX Color Picker Bug

I like to assign one of the buttons on my pen to the color picker tool (Option modifier on Mac). It works fine in other programs like Photoshop, but in Pyxel Edit, my hotkey button doesn't do anything so I have to press the actual keyboard key. I have the latest drivers, and it also works fine in the Windows version of Pyxel Edit so I'm a bit puzzled. Has anyone else experienced this?
